31-year-old Cooper has spent 10 years at Stradey Park and made 113 appearances. He is excited about joining head coach Kevin George and backs coach Paul Fisher at Llanelli RFC.
"I'm delighted my first step into coaching is with Llanelli," said Cooper. "I can't wait to get started."
George said: "With the move to a stunning new stadium imminent, these are exciting times. It's crucial that our premiership players are given every chance to develop as professionals and internationals.
"As part of a superb new coaching structure, I'm determined to play a major role in that process."
Llanelli head of rugby Anthony Buchanan said: "Kevin George is a gifted coach and the addition of Vernon and Paul can only be good for us - they're top quality individuals who have hugely impressive rugby knowledge and experience.
"Llanelli RFC should be a force within Welsh rugby and should be developing players for the region and for Wales. We can only do that with quality coaches."
